Expression of Erwinia chrysanthemi genes coding pectate lyases ininfected potato tissue

Erwinia chrysanthemi mutants, containing transcriptional fusion of one ofthe pectate lyase genes (pelA, pelB, peIC, pelD, pelE, pell, pelL, pelZ) with the reporter gene encoding p-giucuronidase activity, were studied for their ability to cause disease symptoms and to synthesise pectinases after inoculation of potatotubers. The strains affected in pell and pelL genes displayed a reduced virulenceon potato tubers demonstrating the important role of these isoenzymes in softroot disease. Analysis of the bacterial population showed an active multiplication of bacteria during the infection. Similar kinetics of growth were observedfor all mutants and for the wild type strain. Comparison of the mutants ?.nd thewild type strain showed that the pell, pelL and pelZ mutants synthesised reducedlevels of Pels. The expression ofpelA, pelE and pelZ is 5-fold higher in planta thanin in vitro. In contrast, both pell and pelL are highly (ł0-fold factor) induced inplanta, which is characteristic of the plant-inducible pectate lyases
