Application of molecular biology methods for blood testing

In this paper a simple, rapid and reliable test for sex diagnosis has been described. The test is basedon detection of alphoid satellite sequences in undigested genomic DNA using the polymerase chain reaction (PCR) and dried blood specimens serving as the source of DNA. When female DNA was used as atemplate for the reaction, only the expected X-chromosome-specific fragment was detected, while withmale DNA both X-specific and Y-specific-fragments were detected. The Y-chromosome-specific fragment was further characterized by restriction enzyme analysis. The Y fragment was detectable when DNAobtained from an equivalent of 5 n\ of spotted blood was used in the reaction. This test may find applications in sexing of human DNA for forensic purposes, in following host versus graft cells in patients withbone marrow transplants from the opposite sex or in searching for XO/XY mosaicism in patients withTurner syndrome.
