Muscle segment examination in light & electron microscopy

Abstract:

An 26-year-old patient with suspected myopathy was examined. A biopsy of quadriceps sinisterwas performed. Ultrastructural assessment showed morphological changes within muscle fibers andglycogen storage under sarcolemma and in the interfibrillary space (Figs. 1-7). Glycogen was alsopresent in mitochondria (Fig. 8).Analysis of muscle fibers in PAS and PAS staining diastase showed the accumulation of PASpositive substance corresponding to glycogen. Muscle phosphorylase activity is significantlyreduced. These data allow the diagnosis of type V glycogenosis.
