Gene cloning using subtractive hybridization

Abstract:

Genes expressed in temporally, environmentally or tissue specific way can be cloned using subtractivehybridization. It involves hybridization of single stranded cDNA from one cell type with mollar excess ofpolyA+RNAfrom another cell type. Unhybridized, cell specific cDNA is subsequently used or as subtractiveprobe or as material for construction of subtractive cDNA library. Modifications of this basic strategy arealso discussed.
