Abstract:

The metabolism of glucose and glycogen in homogenous porcine liver during perfusion was studied. The liver was preserved according to authors own technique. During 3-hours perfusion the small amount of glycogen (previously changed into glucose) was eluted and caused the increase of glycaemia of perfusing Ringer’s fluid. In the first two hours after connexion of perfused liver to donorpig liver the liberation of almost all of the glycogen stored was confirmed by the histochemical investigation of liver material obtained by oligobiopsy and stained with PAS method. In the same time the increase of glucose and glycogen concentrations in venous blood donor-pig were observed. In the liver of donor-pig in which the blood vesels were closed there was decrease of glycogen to 4% of instial value (100%). The glycogen concentration in straited muscles was constant during perfusion. Performed in the last hour of perfusion bromsulftalein test shown 28% of dye retention Addition of potassium chloride and insulin to the intravenous glucose infusion resulted in only very small deficit of glycogen in perfused liver. This fract was confirmed by histochemical examination. The bromsulftalein te st performed after 45 minutes in donorpig shiwn only 18° о eye retention.
